Over the last few years, there has been a growingneed to advance the use of thermal ablation therapyas a minimally invasive technique in cancermanagement. Consequently, this work was done toinvestigate the use of hyperthermia and hypothermiaby developing economical probes that will effectively destroy volumes of pathologicaltumours. While work on the cryotherapy probe is stillon-going, the RF thermal probe was tested ondifferent tissues and evaluated for effectivetissue destruction capability. The results show that,at power settings of 20 watts for soft tissue, 13watts for kidney tissue, 6 watts for liver tissue, 3watts for brain tissue and 2 watts for lung tissue,the RF thermal probe effectively destroys volumes ofpathological tumours within a temperature range of 50to 100 degrees centigrade during a 15-minute treatment time. This work should help shed more light on the two energy-based treatment methods investigated, and should be of particular interest to interventional radiologists, researchers in the field of thermal ablation therapy, and cancer patients who are considering the use of energy-based therapy as alternative treatment methods.
This dissertation presents a novel technique ofdelivering membrane impermeable molecules into singleliving cells with the assistance of femtosecond (fs)near-infrared (NIR) laser pulses. This techniquepromises several major advantages, namely, very hightransfection efficiency, high cell survival rate( 100%) and fully preserved cell viabilities. In this work, the system for fs NIR opto-injectionwas designed and built. Successful fs NIRopto-injection has been performed on several cellsystems including single mammalian cells, marineanimal eggs, and human cancer cells cultured in atissue-like environment. The connections betweenlaser parameters and cell responses were explored.Dye uptake rate of the target cells was observed todepend on incident laser intensity. Pore size wasfound dependent on incident laser intensity. Theconclusion was made that laser-induced breakdown andplasma-induced ablation in cell membrane are thephysical principles that govern the process of fs NIRopto-injection.
Over the last decades, applications of irreversible electroporation (IRE) have emerged in biotechnology, food industry, and medicine. IRE uses a series of microsecond electric pulses that permanently open cell membranes and lead to cell death. The goal of this book is to address certain aspects of two major IRE applications: disinfection and tissue ablation in the treatment of diseases such as cancer. In relation to disinfection, I discuss IRE for sterilization of drugs, and using IRE for storage of perishable products. In relation to tissue ablation, I present a statistical model IRE treatment planning, and a new method for real-time control of IRE procedure. During the research on the real-time control of IRE in tissues, we realized that it has value beyond the original intended application - it may be used for generation of low cost electric energy. Theoretical analyses and emerging application discussed in this book should be especially useful to professionals in the Biomedical and Food Engineering fields, or anyone who is interested in the field of bioelectricity.
